How Does SUMAtriptan Work?

Sumatriptan is a 5-HT1 receptor agonist, or 'triptan,' which functions as a vasoconstrictor. It achieves its therapeutic effects by binding to 5-HT1B/1D receptors located on intracranial blood vessels and sensory nerves of the trigeminal system.

Common Dosages

  • Tablets: 25mg, 50mg, 100mg
  • Nasal Spray: 5mg, 20mg
  • Injection: 4mg, 6mg


For FDA-approved indications:

  • Migraine or cluster headache: 
  • Oral: 25 to 100mg, may repeat after 2 hours if needed; maximum 200mg/day. 
  • Injection: 1 to 6mg under the skin, repeated once after one hour if needed; maximum 12mg/day. 
  • Nasal spray: One 5mg or 20mg spray in one nostril, repeated once after two hours if needed; maximum 40mg/day.

For off-label uses:

  • Cyclic vomiting syndrome: 6 mg subcutaneously, may repeat after 1 hour in patients with no or partial response. Max 6 doses/week.

  • People who have had an allergic reaction to sumatriptan or other triptans.

  • Not advised to use within 24 hours of another triptan or ergot-type medication.
  • Be careful if you're on monoamine oxidase-A inhibitors (MAOIs), selective serotonin reuptake inhibitors (SSRIs), or serotonin and norepinephrine reuptake inhibitors (SNRIs) due to the risk of serotonin syndrome.
